Project Description:
Single particle electron cryo-microscopy and the associated image processing and 3-D reconstruction technique, collectively known as cryo-EM, has emerged in recent years as a powerful new method that allows biologists to study dynamic biological processes, such as virus assembly and protein translation by ribosome. In cryo-EM, the purified macromolecular particles are frozen and imaged in high-end electron microscope as 2-D projection images at unknown random views. Computational processing of the images is essential to determine the relative views of the different images termed 2-D alignment, and to reconstruct the 3-D structure of the structures by coherently merging large number of the 2-D images based on theory/method similar to that of medical CT scan. This project will develop computational methods for high throughput image acquisition, processing, and 3-D reconstruction.
Expected Student Contributions:
The intern will implement graphic interfaces to existing command line image processing programs, first for desktop computers and then as web service using Purdue DiaGrid. The intern will also research and implement ways of improving the underlying image process algorithms for higher quality 3-D reconstructions. The intern will attend weekly lab meeting and then make a presentation in the lab meeting at the end of the semester.
